Auch im Sanctus und im Agnus Dei werden diese Gelassenheit und Beherrschung erreicht; in verschiedenerlei Hinsicht sind es jedoch das Gloria und das Credo, die das Werk wirklich herausragend machen. Ohne dass jemals der dynamische und nach vorn gerichtete Antrieb verloren ginge, gelingt es Palestrina, dass jedes einzelne Wort des Textes klar und deutlich verstanden werden kann.

It is written for four voices, soprano, alto, tenor, and bass. The Benedictus, however, is for three voices, while the second Agnus Dei is for five. An important characteristic of Palestrina expressed clearly in this work is the use of frequent tempo changes, which are governed entirely by the changing meaning and significance of the text. Agnus Dei The Agnus Dei returns to the imitative polyphony of the Kyrie (the opening of Agnus Dei I repeats that of the Kyrie). As was frequently done in the 16th century, Palestrina adds an extra voice in Agnus Dei II, making seven for this movement, in which is embedded a three-part canon that begins with the head-motive.
